We Can Just Guess, So Here We Start Discussion. My View I think In Next we Will See Fight B/W Tsunade and Pein. As We Know Kishi has Said One of Akatsuki will be brother or sister of hokage, (Interview) so, I think before fight ther ewill be talk b/w tsunade and pein... and we will come to know pein is HER Brother. And as we saw in previous chapter Tsunade already know this guy Pein.
